Analysis of Anticancer Drug induced Cardiotoxicity using a Transgenic Zebrafish Line MieKomachi009
英文抄録本文:
Drug-induced cardiotoxicity is emerging as an important issue among cancer survivors. Especially, anthracyclines and drugs targeting tyrosine kinases have been recognized to carry an unwanted effect on the cardiovascular system. Although a number of efforts have been directed towards prediction of risk,so far no consensus exists on the strategies to prevent and monitor chemoth erapy-related cardiotoxicity. Therefore, it is important to develop an animal model for the assessment of drug-induced cardiotoxicity. Zebrafish are well-suited for use in chemical screens. Small molecules added to the aquatic environment in which they live, are absorbed into the fish without the need for invasive and time-consuming injection. In this study, we used a transgenic zebrafish line(Miekomachi009) in which GFP selectively expressed in heart.Miekomachi009 were exposed to anticancer drug from 4 days post fertilization (dpf) to 7 dpf and analyzed for the cardiac function by fluorescence time-lapse images of the ventricle. We were able to detect significant effects of anticancer drug on the cardiac function which were consistent to anticancer drug induced cardiotoxicity in mammals. This method can be applied to assess cardiotoxicity induced by anticancer drugs.
